    Neuroprotective Effect of Mailuoning on MCAO Rats and Its Antioxidant Mechanism

    Objective:To investigate the antioxidant mechanism of mailuoning on MCAO rats. Methods:18 adult SD rats were randomly divided into control group,MCAO group and mailuoning group,6 rats in each group. A rat middle cerebral artery occlusion (MCAO) model was established by suture method,ischemic 2 h and reperfusion 24 h. 2% 2, 3,5-triphenyltetrazolium chloride (TTC) was used to measure the cerebral infarct volume in rat. The content of reactive oxygen species (ROS) were determined by fluorescence spectrometry. The contents of malondialdehyde (MDA) and glutathione (GSH) in brain tissue were determined by absorbance method. The expression of cysteinyl aspartate specific proteinase (caspase-3),B-cell lymphoma-2 associated X protein (Bax),B-cell lymphoma-2 (Bcl-2) were measured by Western Blot. Results:Mailuoning could significantly decrease the neurological function of MCAO rats and decrease the volume of cerebral infarction. Mailuoning could inhibit the production of ROS and the formation of MDA,meanwhile increase the level of GSH in brain tissue. Mailuoning could also inhibit the expression of caspase-3,Bax,however,increase the expression of Bcl-2. Conclusion:The protective effect of mailuoning on MCAO rats may be related to its inhibition of oxidative stress and apoptosis.

    Depression is a common recurrent psychiatric disorder,which has severely affected quality of human life. So far,the pathogenesis of depression is still unknown and awaits clarification. This paper is a summary of the pathophysiology and animal models of depression,and provides reference for the treatment of depression and the evaluation of antidepressant drugs.

    Blood-cerebrospinal fluid barrier hinders the treatment of brain diseases,while targeted agents can not only selectively concentrate on the foci,but also increases the concentration of drugs and thereby reducing the toxicity and side effects,which popularly directs the treatment of brain diseases nowadays. This paper summarizes several brain target ligands applied broadly and their delivery strategies over the last five years,involving drug-carrying nanoparticles that transported to the brain by means of receptor-mediated,vector-mediated,and trans-cell-swallowing,which makes it possible to accumulate drugs in the brain,and needs to be prospectively researched and developed.

    The Mechanism of Dexmedetomidine and Its Application in Clinical Anesthesia

    Alpha 2- adrenergic receptor agonist has sedative,anti-anxiety,hypnotic and analgesic effects. Dexmedetomidine (Dex) is a new,potent and highly selective alpha 2- adrenergic receptor agonist.The present study showed that Dex could reduce the sedative and opioid dose,stabilize hemodynamics,and had the effects on anti-inflammation and organ protection,anti-shaking and salivation as well as other pharmacological properties in a wide range of clinical applications. Dex is considered to be a safe and effective adjuvant,or in single use of therapeutic agent in clinical anesthesia,it has a broad application prospects. From the mechanism of Dex,preoperative,intraoperative nerve block and spinal anesthesia,general anesthesia and postoperative adjuvant drug use,organ protection,ICU sedation etc. This article reviews its application in clinical anesthesia,and provide a theoretical basis for the clinical application and basic research of Dex.

    It is known that atherosclerosis is the pathological basis of ischemic cerebral apoplexy. A large number of studies suggest that plasma lipoprotein-associated phospholipase A2(Lp-PLA2), as a new inflammation factor,plays a role in the development of atherosclerosis, and it is a independent risk factor for cardiovascular and cerebrovascular disease. This review summarizes the relationship between Lp-PLA2 and the ischemic stroke.

    Study on the Neuroprotective Effects of Phytoestrogens in Alzheimer’s Disease

    Phytoestrogen (PE) is a type of plant component that is widely found in traditional Chinese medicine and has similar bioactivity of estrogen. It has been widely acknowledged about the role of phytoestrogens in the prevention and treatment of cardiovascular diseases,osteoporosis, tumors,and improvement postmenopausal syndrome in women. In recent years,more and more studies have shown that phytoestrogens play a protective role in the prevention and treatment of Alzheimer’s disease. Because they are widely derived from plants,and have no obvious side effects,concomitantly can be used as preventive health products,making phytoestrogens have a wide range of applications and prospects. This text reviews the source,classification,biological activity of phytoestrogens,and their pharmacological effects on improving learning and memory function, reducing the production of β-amyloid,antagonize the toxicity of β-amyloid and enhancing the function of cholinergic neurons ,the anti-oxidative stress and the neuroprotective effect in eliminating free radicals in Alzheimer’s disease,as well as the mechanism of phytoestrogen exerting neuroprotective effect through direct genomics and non-genomic effects. It provides new ideas and strategies for the development of Alzheimer’s disease prevention and treatment drugs making it a promising alternative to estrogen.

    Excellular senile plaques deposits and intracelllar neurofibrillary tangles are the typical neuropathologies of Alzheimer’s disease (AD). AD is also characterized by cholinergic hypofunction in hippocampus caused by basal forebrain cholinergic neurons( BFCNs) dysfunction. This review focues on ACh neurotransmission,BFCNs projective system,the regulatory roles of BFCNs in cognition and the progression of BFCNs dysfunction in AD.
