    Experimental Study on Pharmacodynamics of Wuwei Depileser in Treating Hemorrhoids

    Objective: To study the multiple effects including anti-inflammation, analgesia, facilitation of ulcer healing of Wuwei depileser. Methods: The anti-inflammation effects of Wuwei depileser were observed using the toe swelling model in rats, ear swelling model in mice, and the mouse peritoneal capillary permeability increase model induced by acetic acid. The analgesia effects of Wuwei depileser were observed using hot plate and acetic acid stimulation. The facilitation of ulcer healing of Wuwei depileser were observed using the model of rat perianal ulcers induced by acetic acid. Results: Wuwei depileser has a significant anti-inflammatory effect on toe swelling model in rats and ear swelling model in mice, and inhibits acetic acid-induced increase in permeability of peritoneal capillaries. It also markedly suppresses pain caused by hot plate and acetic acid stimulation, facilitates the healing of perianal ulcers and swelling. Conclusion: These results indicate that Wuwei depileser has multiple effects including anti-inflammation, analgesia, facilitation of ulcer healing.

    Prediction of the Target of Quercetin in the Treatment of Breast Cancer Based on Network Pharmacology

    Objective: To predict the potential target of quercetin (Que) in the treatment of breast cancer (BC) through network pharmacological prediction. Methods: The target of Que was predicted through the SwissTargetPrediction database; BC targets were obtained through TTD database. The common targets intersection of Que and BC was obtained by mapping the target of Que and BC. The protein-protein interaction (PPI) network was constructed and analyzed through STRING database; DAVID database was used for Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. Molecular docking and analysis of some common targets of quercetin-BC analyzed above. Results: 77 targets and 118 targets were selected from Que and BC, and 20 common targets were obtained from the intersection. After the protein-protein interaction (PPI) network was constructed, the post-topological PPI network composed of 13 common targets was obtained through topology. Through GO enrichment analysis, 67 biological processes, 10 cell components and 27 molecular functions were obtained. KEGG pathway analysis obtained 20 pathways. Through PPI, GO and KEGG analysis, five potential targets (KDR, AKT1, IGF1R, INSR, EGFR) of BC and Que were obtained. Molecular docking verification was performed on some of the targets, and it was found that the docking energy was low, which was consistent with the results of PPI, GO and KEGG analysis. Conclusion: The potential target of Que in the treatment of BC has been found by using network pharmacology, which provides a new theoretical idea for the follow-up treatment of BC with quercetin.

    To Explore the Mechanism of Action and Active Substances of Panax Notoginseng in the Treatment of Gastric Cancer Based on Network Pharmacology and Molecular Docking

    Objective: Through network pharmacology and molecular docking technology, the only known plant component of Babaodan in the treatment of gastric cancer potential active ingredients and its mechanism of action. Methods: The active ingredients and related target proteins of Panax Notoginseng were screened by TCMSP and other databases, and the gastric cancer related targets and drug target proteins mined by GeneCards and DisGeNET databases were intersected. Cytoscape3.7.0 was used to analyze pharmacodynamic components to establish an effective component-target-disease relationship model. The PPI protein interaction map of intersecting targets was constructed by using String database. The enrichment analysis of KEGG pathway and GO classification was carried out using David database. AutoDock Tools 1.5.6 software was used to verify the molecular docking of key active ingredients and core targets of Panax Notoginseng, and PyMol 2.5 software was used to draw the docking model diagram. Results: 99 active ingredients of Panax Notoginseng were selected and 273 target genes were mapped. Through GeneCards database, 15118 gastric cancer disease genes and 234 intersection targets of Notochi and gastric cancer were collected, and 165 KEGG enrichment pathways and 195 GO functional enrichment items were obtained, mainly including cancer pathway, PI3K-Akt pathway, MAPK pathway and Ras pathway. And RNA polymerase promoter transcription positive regulation, signal transduction, gene expression positive regulation, positive regulation of cell proliferation, negative regulation of cell apoptosis and other biological processes; Among them, stigasterol and ginsenoside rh2 with high Hub value had strong molecular docking relationship with gastric cancer genes IL6, TP53 and TNF. Conclusion: Panax Notoginseng may play a role in the treatment of gastric cancer through regulating cancer pathways, PI3K/Akt pathway, MAPK pathway and Ras signaling pathway, among which stigmosterol and ginsenoside rh2 may be one of the key active ingredients of Panax notoginseng.

    Preliminary Study on the Potential Bioinformatics Mechanism of the Occurrence and Development of Hypertensive Disorder Complication Pregnancy

    Objective: To explore the potential mechanism affecting the occurrence and development of hypertensive disorder complication pregnancy based on bioinformatics. Methods: Two datasets of GSE190639 and GSE206763 in GEO database were selected, and differential genes and differential miRNAs were screened by GEO2 R online platform, respectively. Volcano map, PPI map and KEGG enrichment map were drawn and analyzed accordingly. A total of 80 patients with pregnancy-induced hypertension (case group) and 80 healthy pregnant patients (control group) who were treated in the Second Affiliated Hospital of Hebei North University from January 2022 to December 2022 were selected. Immunohistochemistry was used to detect the expression of hypoxia-inducible factor-1ɑ and miR-34ɑ-5p in placental tissues of the two groups. Results: A total of 42 differential genes were screened from the GSE190639 data set, including 31 up-regulated genes and 11 down-regulated genes. PPI diagram showed that there were 21 genes related to up-regulated genes, which were mainly enriched in signal pathways such as hypoxia inducible factor-1. There were 81 differential miRNAs in the GSE206763 data set, and the up-regulated miRNAs were miR-518b, miR-29b-3p, miR-323b-5p, miR-34ɑ-5p, miR-601, etc. The down-regulated miRNAs included miR-25-3p, miR-150-3p, miR-125b-5p, etc., which were mostly enriched in signaling pathways such as hypoxia inducible factor-1. Compared with the control group, hypoxia-inducible factor-1ɑ and miR-34ɑ-5p were significantly increased in the immunohistochemical case group (P<0.01). Pearson analysis showed that there was a positive correlation between the two (r=0.265, P< 0.05). Conclusion: miR-34ɑ-5p may be involved in the occurrence and development of hypertensive disorder complication pregnancy by regulating the expression of HIF-1ɑ.

    Musculoskeletal Ultrasound Evaluation of A Clinical Study of TGP in the Treatment of Rheumatoid Arthritis

    Objective: To investigate the clinical effect of TGP on bone destruction in patients with Rheumatoid Arthritis (RA) by musculoskeletal ultrasound. Methods: A total of 56 patients with rheumatoid arthritis who were admitted to our hospital from 2021-07 to 2023-07 and were randomly divided into the treatment group and the control group with 28 patients in each group according to the random number table method. The control group was treated with methotrexate, and the treatment group was treated with methotrexate and TGP for 12 weeks. Inflammatory indicators such as ESR and CRP were observed before and after treatment. The Clinical effect of the two groups was evaluated by ACR20/50/70. Musculoskeletal ultrasound was used to detect synovial, the synovium blood flow signal, bone erosion and joint effusion. The above indexes were compared between the two groups before and after treatment. Results: ① The control rate of ACR20/50/70 in the treatment group was higher than that in the control group, and the inflammatory indexes ESR and CRP in both groups were lower than before treatment, but the treatment group was significantly higher than the control group. ② Compared with before treatment, synovial, articular effusion and synovial blood flow were all decreased in 2 groups, and the decrease was more obvious in the treatment group, the difference was statistically significant, while there was no statistically significant difference in bone destruction before and after treatment. Conclusion:TGP can improve the standard reaching rate, reduce the symptoms of patients with rheumatoid arthritis in the acute phase, inhibit inflammatory response, protect joints, prevent RA bone damage, and delay the progression of rheumatoid arthritis.

    Objective: To Explore the clinical characteristics of Huntington's disease patients, especially mental disorders, and improve the diagnosis rate of HD diseases based on understanding of psychiatric symptoms. Methods: In January 2022, under the principle of patients informed and voluntary, the clinical data of one HD patient receiving a section of the first hospital affiliated with the first hospital, Hebei Northern College, will be collected, revisited and followed up. Results: Two of the patient's family have psychiatric symptoms such as physical involuntary movement, walking instability, irritation and anxiety, this forensic witness sudden mental disorder, its CGA copy number is 43 times, HAMA score: 16, HAMD score: 18, through the relevant literature review to learn that psychiatric symptoms may be 10 years earlier than motor symptoms. Conclusion: Early diagnosis of HD is very difficult, of which family history is important, early psychiatric symptoms, imaging and pathology are equally important, and early diagnosis of HD patients is of great significance to early prevention, improvement of prognosis and improvement of patients' quality of life.

    Study on the Effect of Compound Lactic Acid Bacteria Adjuvant Regimen in the Treatment of Helicobacter Pylori Infection

    Objective: To investigate the clinical study of compound Lactobacillus assisted high-dose dual regimen in the treatment of adult Helicobacter pylori (Hp) patients. Methods: 300 patients diagnosed with Hp infection in the outpatient department of Nanping First Hospital Affiliated to Fujian Medical University were randomly divided into experimental group (n=150) and control group (n=150). The experimental group was treated with high-dose dual standard regimen of Venolazan, amoxicillin and complex lactic acid bacteria. The control group was treated with high dose dual standard regimen of Venolazan and amoxicillin alone. The treatment course of the two groups was 10 days, and the eradication rate, effective rate and incidence of adverse reactions of anti-HP treatment in the two groups were evaluated. Results: The Hp eradication rate of 13C breath test, the healing of gastroscopy ulcer, the improvement of clinical symptoms of chronic gastritis and the incidence of adverse drug reactions were recorded more than 4 weeks after the end of treatment course (P>0.05). There was no statistically significant difference between the two groups (P>0.05), while the incidence of adverse drug reactions was statistically significant (P<0.05). Conclusion: The combination of compound lactic acid bacteria and high-dose combination therapy for the treatment of Hp infection may not improve the eradication rate of Hp, but it can reduce the incidence of adverse reactions to antibiotics. The variety of medication used is small, easy for patients to accept, and provides compliance, which is worthy of clinical development.

    Obesity-associated chronic low-grade inflammation is a major driver of metabolic disorders in the body, which is involved in the onset and progression of metabolic diseases such as metabolic syndrome and insulin resistance, and for which there is currently no effective treatment, an important reason being that the mechanisms of inflammation regulation are not fully understood. Vagal-mediated cholinergic signaling can improve obesity-related inflammation by regulating the body's metabolic homeostasis and immune function. Recent studies have shown that the imbalance between the pro-inflammatory and anti-inflammatory systems due to the impairment of the cholinergic anti-inflammatory pathway, which is the basic anti-inflammatory pathway in obesity, is the key to developing inflammation. Metformin (MF) is a clinically useful hypoglycaemic drug whose mechanism of action has been elucidated to control inflammation and improve metabolic disorders. It has been found that MF exerts synergistic effects with the activation of cholinergic signaling pathways in the body, and there may be an important link between the two, which makes it possible for MF to exert its effects by targeting cholinergic anti-inflammatory pathways. In this paper, we would like to review the high-fat diet-induced impairment of cholinergic anti-inflammatory pathways and the possible mechanisms of MF targeting cholinergic anti-inflammatory pathways, to provid new insights for the prevention and treatment of obesity-driven inflammatory and metabolic diseases.

    Platelet-rich plasma (PRP) is a platelet plasma concentrate containing mainly high concentrations of platelets, leukocytes and fibrin, obtained by centrifugation of autologous blood, which is activated to release large amounts of cell growth factors (GFs) and cytokines, and has been shown to be effective in reducing inflammatory responses, promoting tissue anabolic capacity and having a strong regenerative potential. The abundance of growth factors and cytokines plays an important role in the initiation and regulation of the soft tissue regenerative microenvironment. Peripheral nerve injuries, bone defects, muscle ligament injuries, articular cartilage disease and chronic non-healing wounds caused by trauma, sports-related or other types of trauma are commonly encountered in orthopaedic clinical practice and often require surgical treatment. This article reviews the recent use of PRP in the treatment of orthopaedic injury-related disorders and discusses future research on the use of PRP in the treatment of orthopaedic injury-related disorders and provides a reference for clinical treatment.
