Objective:To explore the effect of sea buckthorn oil on cell proliferation and apoptosis of human gastric cancer cells HGC-27. Methods:HGC-27 cells were treated with sea buckthorn oil. The cell viability was measured by CCK-8 kit. Western blotting was used to detect the protein expression of p53,p21 and Bcl-2. Results:Cell viability tests results showed that there is no effect on HGC-27 cell viability when DMSO content in cell culture medium is less than or equal to 0.1%. HGC-27 cells were treated with sea buckthorn oil dissolved in DMSO. The proliferation of HGC-27 cells was inhibited by sea buckthorn oil at the concentration of 5,10,20,
40 mol·L-1(P<0.05). Compared with control group,the expression level of p53 in three different concentration of sea buckthorn oil (10,20,40 mol·L-1) group were significantly increased(P<0.05). The expression level of Bcl-2 was significantly decreased and p21 was significantly increased in the concentration of sea buckthorn oil (5,10,20,40 mol·L-1) group. Conclusion:Sea buckthorn oil can inhibit HGC-27 proliferation and promote apoptosis by p53 signaling pathway,and has anti-cancer effect.
