Progress on Research of 5-Lipoxygenase in Alzheimer’s Disease

Abstract: 5-lipoxygenase plays a key role in catalyzing the conversion of arachidonic acid (AA) to 5-hydroperoxy-6,8,11,14-eicosatetraenoic acid (5-HPETE),which can then be metabolized into different leukotrienes (LTs). The recently studies have shown that 5-lipoxygenase is involved in the pathophysiology of Alzheimer’s disease. Inhibition of 5-lipoxygenase decreases amyloid β peptide (Aβ) and tau hyperphosphorylation,mitigates neuroinflammation,regulates synaptic function and ameliorates cognitive disorders in Alzheimer’s disease. In this review,we will summarize studies of 5-lipoxygenase in Alzheimer’s disease pathogenesis.
