    Study on Association of CD14 -159C/T and Cardiovascular Disease in the Han Nationality from Zhangjiakou

    Objective:To investigate the correlation between the physicochemical properties,functions and gene polymorphisms of the protein encoded by the cluster of differentiation antigen 14(CD14) gene and cardiovascular disease,so as to provide basis for the prevention and treatment of cardiovascular disease. Methods:Plasma samples were collected from 75 patients with cardiovascular disease (including 42 male patients and 33 female patients),and 125 healthy controls (including 80 male patients and 45 female patients). Genomic DNA was extracted from their venous blood. The polymorphism of - 159C / T (rs2569190) locus of CD14 gene was detected by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP),and the results of enzyme digestion were observed by electrophoresis. The correlation between the polymorphism of-159C/T and cardiovascular disease was detected by genotype frequency. The physical and chemical properties,structure,function and interaction of CD14 gene coding protein were analyzed by bioinformatics methods to demonstrate the relationship between CD14 gene coding protein and cardiovascular disease. Results:The distribution frequency of TT genotype of CD14-159C / T(rs2569190) in the diseased group was 56%,which was significantly higher than that in the normal control group (40.8%). The difference was statistically significant( P<0.05),and the other genotypes were not statistically significant (P>0.05). CD14 is a secretory protein,located in the extracellular matrix,without transmembrane region and weak hydrophilicity. There were more irregular curls in the secondary structure of CD14 protein (41.87%). CD14 interacts directly with 10 proteins and plays an important role in Toll like receptor 4 signaling pathway and MyD88 dependent toll like receptor signaling pathway. Conclusion:The-159C/T( rs2569190) TT genotype of CD14 gene is closely related to the occurrence of cardiovascular disease. CD14 protein may play an important role in the occurrence and development of cardiovascular disease.

    The Regulation of SP/NK1 Receptor on Gastrointestinal Motility in Experimental Gastric Ulcer Rats

    Objective:To investigate the effect of substance P( SP)/ NK1 receptor pathway on contractile function of isolated pyloric smooth muscle strips by detection gastrointestinal function and SP expression during experimental gastric ulcer. Methods:62 SD rats were divided into normal group(n=6),saline control group (n=24) and ulcer group (n=32). Gastric ulcer model were used acetic acid-induced in rats,and the ulcer area was measured. Gastric emptying rate and intestinal propulsive function were observed by intragastric administration of methyl cellulose. The effects of SP and NK1 recepter antagonist on the contractile reactivity of isolated pyloric annular smooth muscle strips with a tension transducer and a multi-channel physiological signal acquisition and processing system. Results:The typical “Ring dike sign”was formed on the 4th day,the ulcer gradually healed from the 10th to 14th day,and the scar tissue was formed on the 28th day. Immunohistochemical results showed that SP positive cells were mainly distributed in fundic gland cells,submucosa and myometrium of gastric mucosa. The integral optical density of SP in ulcer at the 4th,10th and 14th days was significantly lower than that in normal and control groups (P<0.05,P<0.01). Gastric emptying rate in ulcer group was significantly lower than that in normal group and saline group (P<0.05) . The contractile amplitude of pyloric annular muscle strips in ulcer group was decreased in different degrees compared with that in normal group (P<0.01) ,when NK1 receptor antagonist was added,the contraction amplitude and frequency decreased significantly(P<0.05,P<0.01). Conclusion:The mechanism of decreased gastrointestinal motility in gastric ulcer is related to SP/NK1 receptor signal pathway.

    Prediction of Core Targets of Umbilical Cord Mesenchymal Stem Cells for Breast Cancer Treatment

    Objective:Using network databases and bioinformatics technology platforms,analyze the data of Umbilical cord mesenchymal stem cells (UC-MSCs) in the treatment of breast cancer (BC),and predict the core target of UC-MSCs in the treatment of BC. Methods:Use the GEO database in NCBI (www.ncbi.com.cn) to search the original data set GSE64827 of UC-MSCs treatment of BC. Then use R software to standardize the data to obtain differential genes;use STRING database to perform protein-protein interaction function network analysis and topology analysis,use DAVID online tools and R software to carry out GO enrichment analysis and KEGG pathway analysis of related genes. Results:Using R software to screen out 11 differential genes,do network analysis and topological analysis of their protein-protein interactions (relevant genes with confidence greater than or equal to 0.7),suggesting the association degree of ALDOC,IGFBP3,DDIT4 and PFKFB4 is higher. GO analysis and annotation obtained 3 biological processes,namely fructose metabolism process,glycolysis process and hypoxia reaction process. The main genes involved are PFKFB4,ALDOC,DDIT4 and CA9. KEGG pathway analysis yielded a fructose and mannose metabolic pathway,involving genes ALDOC and PFKFB4. Conclusion:This study uses network databases and bioinformatics technology platforms to predict that the core targets of UC-MSCs for breast cancer treatment are ALDOC,PFKFB4 and DDIT4.

    Effect of Acupuncture Combined with Traditional Chinese Medicine in Treatment of Diabetic Retinopathy:A Meta-Analysis

    Objective:Meta-analysis was conducted to investigate the effect of acupuncture combined with Traditional Chinese medicine (TCM) in the treatment of diabetic retinopathy(DR). Methods:Studies on the effect of combination treatment in DR in PubMed,Embase,Web of Science,Cochrane Library,CNKI,WanFang and SinoMed database were systematically reviewed and included.The data analysis was performed using Stata software. Results:A total of 10 studies were included. Compared with other treatment,the combination treatment significantly improved the overall effective rate( RR=1.21,95%CI between 1.13 to 1.30),visual acuity level( WMD=0.18,95%CI between 0.11 to 0.25),macular thickness (WMD=-32.25,95%CI between -48.94 to -15.56),and visual acuity (WMD=2.92,95%CI between 2.57 to 3.27). Compared with the control group,the difference was statistically significant (P<0.01). Conclusion:Acupuncture combined with Chinese Herb in the treatment of DR can improve the visual acuity and visual acuity,reduce the thickness of the macula,which slows down disease progression and improve the symptoms of patients.

    Correlation between Right-to-Left Cardiac Shunt and White Matter Lesions in Patients with Migraine

    Objective:To study the correlation between right-to-left shunt (RLS) and white matter lesions (WML) in patients with migraine. Methods:152 patients who were diagnosed as migraine and met the inclusion criteria were selected from the neurology clinic of the First Affiliated Hospital of Hebei North University from September 2020 to November 2021. The general data of patients were recorded and the migraine scale was registered. All patients were examined by contrastenhanced transcranial Doppler (c-TCD) and magnetic resonance imaging (MRI). According to the results of cranial MRI,the patients were divided into WML+ group and WML- group. Spss25.0 software package for statistical analysis to check whether there is correlation between RLS and WML. Results:There were 50 cases of RLS(+) in WML+ group and 25 cases of RLS(+) in WML group.The difference between the two groups was statistically significant( χ2 = 4.016,P = 0.045).The scores of white matter lesions in different grades of right-to-left shunt were statistically significant (Rho=0.208,P=0.010). Conclusion:The incidence of WML in migraine patients is related to RLS. The higher RLS level is,the more serious WML is.

    Analysis of Chemical Composition and Content of Ardisia japonica (Thunb) Blume

    Objective:To study the chemical constituents and determine the content of the chemical constituents in Ardisia japonica (Thunb) Blume. Methods:Separation by silica gel,D101 macroporous adsorption resin,C18 and other chromatographic methods. Purification of preparative chromatograph. Identification of molecular structure by MS,UV,IR,NMR and other spectroscopic methods. The content of compound 2 in Ardisia japonica (Thunb) Blume was determined by HPLC with the homemade compound 2 as the control substance. Results:Four compounds were found from Ardisia japonica (Thunb) Blume,2-methyl-5-[10’(Z)-pentadecenyl]-resorcinol (1),2-methyl-5-[8’(Z)- tridecylene]- resorcinol (2),2-methyl-5-tridecyl-resorcinol (3),2-methyl-5-pentadecyl- resorcinol (4) respectively. The content of compound 2 in roots,stems and leaves of Ardisia japonica (Thunb) Blume was 16.79 mg·g-1、 19.39 mg·g-1 and 24.59 mg·g-1 respectively. Conclusion: Compound 1 was a new compound. Compounds 3 and 4 were first discovered from Ardisia japonica( Thunb) Blume.

    Research Progress of Chrysophanol Nanoparticles

    Chrysophanol,as the main effective component of traditional Chinese medicine rhubarb,Polygonum multiflorum and Polygonum cuspidatum,has a variety of pharmacological effects such as anti-cancer, anti-tumor,neuroprotection,improvement of learning and cognitive function,myocardial protection and so on. However,chrysophanol as a single component has not been independently applied in clinical practice. With the continuous application of nanotechnology in the field of biomedicine,studies on the pharmacological effects of chrysophanol and the application of nano preparations to improve bioavailability have emerged one after another in recent years. In this paper,the research status and existing problems of nano preparations in recent years were discussed,and four systems of nano preparation carrier materials were systematically introduced. The pharmacological action of chrysophanol and the research progress of chrysophanol nano preparation are discussed in detail,in order to provide some research direction and thinking for the nano preparation and chrysophanol nano preparation in future.

    Correlation between CHA2DS2-VASc Score Combined with Neutrophil to Lymphocyte Ratio and No Reflow after Direct PCI in Patients with Acute STEMI

    Objective:To investigate the relationship between CHA2DS2-VASc score combined with neutrophil to lymphocyte ratio (NLR) and no-reflow phenomenon after direct percutaneous coronary intervention (PCI) in patients with acute ST-segment elevation myocardial infarction (STEMI). Methods:A total of 275 patients with acute STEMI who received direct PCI in the Department of Cardiology,Hebei General Hospital from September 1,2018 to December 31,2019 were selected as the study subjects. All clinical data and examination indicators of patients were collected. According to the results of angiography,they were divided into two groups:the reflow group (n=228) and no-reflow group (n=47).The independent influencing factors of no-reflow after PCI in STEMI patients were analyzed,and receiver operating characteristic (ROC) curve was drawn to analyze the predictive value of CHA2DS2-VASc score,NLR and their combined factors for no-reflow after PCI in acute STEMI patients. Results:CHA2DS2-VASc score,NLR and intraoperative predilation in patients no-reflow were higher than those in reflow group (P<0.05). Binary Logistic regression analysis showed that CHA2DS2-VASc score and NLR were independent influencing factors of no-reflow after PCI in acute STEMI patients (P<0.05). ROC curve results showed that:CHA2DS2-VASc score (AUC:0.640,95%CI0.562~0.719,P=0.002),NLR (AUC:0.631,95%CI0.541~0.721,P=0.005) had certain predictive power for no-reflow phenomenon,but the combined predictive factor(AUC:0.675,95%CI0.589~0.762,P<0.001)were higher in AUC,which could better predict the occurrence of no-reflow after direct PCI in STEMI patients. The predictive power of combined predictors was compared with CHA2DS2-VASc score alone and NLR alone. There were no statistical differences(P>0.05). Conclusion:CHA2DS2-VASc score and NLR are independent predictors of no-reflow.The combination of the two has a good predictive value for the occurrence of no-reflow after PCI in acute STEMI patients,but does not show a better predictive value than CHA2DS2-VASc score or NLR alone.

    The occurrence and development of gastric cancer is a multi-step and multi factor complex process,which mainly involves DNA mutation,changes of genetic factors and abnormal changes of various cell signal transduction. Phosphatidylinositol 3-protein kinase (PI3K)/protein kinase B (AKT) signaling pathway is involved in the malignant process of tumor cells. The disorder of this pathway can cause a variety of diseases. In recent years,due to the in-depth study of tumor molecular basis,it is found that the activation of this pathway is closely related to the proliferation,invasion,metastasis,apoptosis and angiogenesis of gastric cancer cells. At present,targeting this signal pathway has become a new trend in antitumor research. The research progress of gastric cancer and PI3K/AKT signal pathway is summarized as follows.

    Progress in the Study of Changes and Mechanisms of Urinary Exosomes in Metabolic Diseases and Cardiovascular Diseases

    Exosomes are membranous vesicles 30-100 nm in diameter that are actively secreted by cells to the outside of the cell. They can be extracted and isolated from urine and other biological fluids (e.g.,plasma,amniotic fluid,saliva,ascites,etc.) and carry proteins,nucleic acids,lipids,etc. that can transmit genetic material between cells and participate in cellular immune regulation. In the physiological state,urinary exosomes are mainly derived from various cells facing the lumen,and the proteins and RNAs contained in them are involved in the exchange of information between cells in the microenvironment of chronic diseases. Recent studies have shown that the extraction and isolation of urinary exosomes can be used as a diagnostic criterion for certain chronic diseases and even to determine prognosis. This article summarizes the progress of research on the therapeutic applications and mechanisms of action of urinary exosomes in metabolic diseases and cardiovascular diseases.
