摘要:

目的:探讨穿龙薯蓣皂苷元诱导胶原性关节炎(collagen induced arthritisCIA)小鼠CD4+CD25-T 细胞向调节性T 细胞(regulatory T cellsTreg)转化的影响。方法:采用免疫磁性细胞分离法(magnetic activated cell sortingMACS)分离CIA 小鼠的CD4+CD25-T 细胞。通过不同浓度(2512.56.25 μmol·L-1)的穿龙薯蓣皂苷元诱导CD4+CD25-T 细胞向Treg 细胞分化,用FCM 检测Treg 细胞比率,Real-Time PCR 法检测Foxp3 mRNA的水平,ELISA 法检测培养上清白细胞介素10interleukin 10IL-10)的含量。结果:免疫磁性细胞分离法分选出的CD4+CD25-T 细胞纯度达93.89%。与对照组相比,穿龙薯蓣皂苷元给药组Treg 细胞比率明显上调(P<0.05),Foxp3 mRNA 的表达水平和培养上清中IL-10 的水平均明显增加(P<0.05)。结论:穿龙薯蓣皂苷元可以诱导

CIA 小鼠CD4+CD25-T 细胞向Treg 细胞分化,并促进Foxp3 的表达。

Abstract:

ObjectiveTo investigate the roles of diosgenin in the differentiation of regulatory T cells from CD4+CD25-T cells in collagen induced arthritis mice. Methods

CD4+CD25-T cells of CIA mice isolated by immunomagnetic beads on magnetic activated cell sorting device were co-cultured with different concentrations of diosgenin2512.56.25 μmol·L-1for 48 hours. The ratio of Treg cells was tested by flow cytometry. The level of Foxp3 mRNA was measured by Real-Time PCR. The level of serum IL-10 was detected by ELISA. ResultsThis study separated CD4+CD25-T cells of 93.89% purity. The ratio of Treg cellsthe level of IL-10 and Foxp3 mRNA were increased significantly in diosgenin groups compared with those in control groupP<0.05. ConclusionDiosgenin may induce the differentiation of CD4+CD25-T cells to regulatory T cells in CIA mice and up-regulate the expression of Foxp3.
