摘要:

目的:探究沙棘油对人胃癌HGC-27 细胞增殖与凋亡的影响。方法:使用沙棘油干预人胃癌细胞HGC-27 细胞,使用CCK-8 试剂盒检测细胞活性、Western Blotting 实验检测凋亡基因P53P21Bcl-2 蛋白表达水平。结果:细胞活性实验显示,小于等于0.1% 助溶剂二甲基亚砜(dimethyl sulfoxideDMSO)细胞培养液对胃癌HGC-2 细胞活性无影响,使用DMSO 溶解沙棘油干预胃癌细胞,当沙棘油浓度为5102040 μg·mL-1 均能够较好的抑制HGC-27 细胞的增殖(P<0.05)。Western Blotting 结果显示,较对照组当沙棘油浓度为102040 μg·mL-1 时,P53 蛋白表达量均明显增加;较对照组当沙棘油浓度为5102040 μg·mL-1 时,Bcl-2 蛋白表达量均明显降低,P21 蛋白表达量均明显升高(P<0.05)。结论:沙棘油可通过P53 信号通路抑制人胃癌细胞增殖,促进胃癌细胞凋亡,达到抗肿瘤作用。

Abstract:

ObjectiveTo explore the effect of sea buckthorn oil on cell proliferation and apoptosis of human gastric cancer cells HGC-27. MethodsHGC-27 cells were treated with sea buckthorn oil. The cell viability was measured by CCK-8 kit. Western blotting was used to detect the protein expression of p53p21 and Bcl-2. ResultsCell viability tests results showed that there is no effect on HGC-27 cell viability when DMSO content in cell culture medium is less than or equal to 0.1%. HGC-27 cells were treated with sea buckthorn oil dissolved in DMSO. The proliferation of HGC-27 cells was inhibited by sea buckthorn oil at the concentration of 51020

40 mol·L-1P<0.05. Compared with control groupthe expression level of p53 in three different concentration of sea buckthorn oil 102040 mol·L-1group were significantly increasedP<0.05. The expression level of Bcl-2 was significantly decreased and p21 was significantly increased in the concentration of sea buckthorn oil 5102040 mol·L-1group. ConclusionSea buckthorn oil can inhibit HGC-27 proliferation and promote apoptosis by p53 signaling pathwayand has anti-cancer effect.
