摘要: 目的:研究马钱苷对神经毒素1- 甲基-4- 苯基-1,2,3,6- 四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)诱导的PC12 细胞自噬的影响。方法:MPTP 作用于PC12 细胞,观察MPTP 在不同时间和不同浓度对PC12 细胞中自噬相关因子微管相关蛋白1 轻链3-β(microtubule-associated protein1light chain 3-β,LC3B)的表达影响,并以此为基础探索马钱苷对MPTP 诱导的细胞自噬的影响和其潜在的作用通路。结果:PC12 细胞经MPTP 处理12 h 后,LC3B 的表达到达最高水平,且MPTP 在100 μg·mL-1 浓度下作用最强,而马钱苷在200 μg·mL-1 的浓度时降低MPTP 诱导的LC3B 高表达,在400 μg·mL-1 的浓度时降低了溶酶体膜相关蛋白2(lysosome associated membrane protein 2,LAMP-2)溶酶体标记蛋白的表达。结论:MPTP 在100 μg·mL-1 浓度下作用12 h,PC12 细胞内自噬蛋白的表达增加。马钱苷可降低MPTP 诱导的PC12 细胞内自噬体水平,并可能通过增强溶酶体对自噬体的消化实现的。

Abstract: Objective:To investigate the effects of loganin on MPTP-induced autophagy in PC12 cells. Methods:The autophagy model was established on PC12 cells by neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine( MPTP). The expression of autophagy-related factor microtubuleassociated protein1light chain 3-β( LC3B) in PC12 cells was investigated at different concentrations and different time after MPTP administration. Then,the effects of loganin on the autophagy related signaling pathway induced by MPTP administration in PC12 cells were detected. Results:LC3B was significantly up-regulated at 12 h after MPTP treatment and the concentration of MPTP at 100 μg·mL-1 showed the strongest action on LC3B expression. Compared to MPTP treated group,the expression of LC3B was significantly down-regulated by loganin at dose of 200 μg·mL-1 combined with MPTP treatment. Furthermore,expression of lysosome associated membrane protein 2 (LAMP-2) was significantly downregulated by loganin at dose of 400 μg·mL-1 combined with MPTP treatment. Conclusions:MPTP at dose of 100 μg·mL-1 could induce the high expression of autophagy protein at 12 h after treatment. Loganin reduced expression of autophagosomes induced by MPTP in PC12 cells,and the inhibitory mechanism may be related to its enhancing digestion of lysosomes to autophagosomes.
