犀角地黄汤对脓毒症大鼠凝血功能的调节机制

Regulation Mechanism of Xijiao Dihuang Decoction on Coagulation Function in Septic Rats

摘要:

目的:探讨犀角地黄汤对脓毒症大鼠凝血功能的调节机制。方法:将90只大鼠按照随机数字表法分成3组, 治疗组、模型组、假手术组各30只。治疗组、模型组以盲肠结扎穿刺法建立脓毒症模型,假手术组轻轻翻动盲肠, 不做结扎穿刺。建模成功后,治疗组给予犀角地黄汤灌胃,模型组和假手术组给予生理盐水灌胃,连续3 d。治 疗组、模型组、假手术组分别于灌胃后24、48、72 h采血,采用ELISA检测血浆中组织因子(tissue factor, TF)、抗 凝血酶-Ⅲ(antithrombin-III, AT-Ⅲ)、活化蛋白C (activated protein C, APC)、组织因子途径抑制物(tissue factor pathway inhibitor, TFPI)、纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor-1, PAI-1)水平。结果:假手 术组在24、48、72 h的TF、AT-Ⅲ、APC、TFPI、PAI-1比较,差异均无统计学意义(P >0.05)。模型组与假手术组比较, 在24、48、72 h的TF、TFPI、PAI-1升高而AT-Ⅲ、APC降低,差异均有统计学意义(P <0.05)。模型组在24、48、72 h 的TF、AT-Ⅲ、APC、TFPI、PAI-1随时间呈进行性改变,差异均有统计学意义(P <0.05)。治疗组与模型组比较,在 24、48、72 h的TF、PAI-1降低而AT-Ⅲ、APC、TFPI升高,差异均有统计学意义(P <0.05)。治疗组在24、48、72 h的 TF、AT-Ⅲ、APC、TFPI、PAI-1随时间呈进行性改变,差异均有统计学意义(P <0.05)。结论:犀角地黄汤调节脓毒 症大鼠凝血功能的机制与TF、PAI-1降低和AT-Ⅲ、APC、TFPI升高有关,但仍需进一步探索。

Abstract:

Objective:To investigate the regulatory mechanism of Xijiao Dihuang decoction on coagulation function in septic rats. Methods: Ninety rats were randomly divided into treatment group, model group and sham operation group, with 30 rats in each group. The sepsis model was established by cecal ligation and puncture in the treatment group and the model group, and the cecum was gently turned without ligation and puncture in the sham-operation group. After successful modeling, the treatment group was given Xijiao Dihuang decoction by gavage, and the model group and sham operation group were given normal saline by gavage for 3 consecutive days. Plasma tissue factor( TF), antithrombin-III( AT-Ⅲ), activated protein C( APC), tissue factor pathway inhibitor (TFPI) and plasminogen activator inhibitor-1 (PAI-1) levels were detected by ELISA AT 24, 48 and 72 hours after gavage in the treatment group, model group and sham operation group. Results: There was no significant difference in TF, AT-Ⅲ, APC, TFPI and PAI-1 between the sham operation group and the control group AT 24, 48 and 72 h (P>0.05). Compared with the sham operation group, TF, TFPI and PAI-1 in the model group increased and AT-Ⅲ and APC decreased AT 24, 48 and 72 h, and the differences were statistically significant (P<0.05). In the model group, TF, AT-Ⅲ, APC, TFPI and PAI-1 showed progressive changes with time AT 24, 48 and 72 h, and the differences were statistically significant (P<0.05). Compared with the model group, TF and PAI-1 in the treatment group decreased, while AT-Ⅲ, APC and TFPI increased AT 24, 48 and 72 h, and the differences were statistically significant (P<0.05). The levels of TF, AT-Ⅲ, APC, TFPI and PAI-1 in the treatment group AT 24, 48 and 72 h showed progressive changes with time, and the differences were statistically significant (P<0.05). Conclusion: The mechanism by which Xijiao Dihuang Decoction regulates coagulation function is related to the decrease of TF and PAI-1 and the increase of AT-Ⅲ, APC, and TFPI in septic rats, but further exploration is needed.
