Thin Layer Identification and Determination of Paeonol Content in Moutan Cortex from Different Origins

Abstract:

Objective: High performance liquid chromatography (HPLC) was used to determine the content of paeonol in Moutan Cortex of different origins, and thin-layer chromatography (TLC) was used to identify the herbs of Moutan Cortex of different origins. Methods: The separation was performed on a G-type thin-layer chromatography silica gel plate with cyclohexane-ethyl acetate-glacial acetic acid (4:1:0.1) as the unfolding agent, and the color was developed by spraying with 2% vanillic acid ethanol sulfate solution; the separation was carried out on a Chevonne C18 column (4.6 mm × 250 mm,5μm) with the elution of methanol-water (45:55 V/V) as the mobile phase, at the flow rate of 1.0 mL·min-1 and the detection wavelength of 274 nm. The mobile phase was methanol-water (45:55 V/V) at a flow rate of 1.0 mL·min-1, the detection wavelength was 274 nm, the column temperature was 30 ℃, and the injection volume was 10 μL. Results: The results showed that the thin-layer identification spots of 21 batches of peony dandelion herbs were clear, and the contents of dandelion were in the range of 2.7%~2.9%, which were higher than the pharmacopoeia standard, of which dandelion in Shandong province was the highest. Conclusion: The quality of peony bark grown in Shandong, Henan and Anhui is good, with good character, small differences between different batches, and uniform and stable quality.
