摘要: 目的:研究二甲基亚砜(dimethyl sulfoxide,DMSO)对SH-SY5Y细胞存活率和氨基酸代谢的影响。方法:将细胞分为正常组和DMSO组。0.0125%浓度的DMSO作用SH-SY5Y细胞不同时间(24,48 h)后,利用MTT法测定细胞存活率;高效液相色谱法测定细胞中氨基酸的含量。 结果:与正常组相比,在0.0125% DMSO作用下,细胞活力和形态无显著差异,但细胞内氨基酸含量变化有显著差异(P<0.05)。其中作用24 h,细胞中有7种氨基酸:谷氨酸(glutamic acid,Glu)、谷氨酰胺(glutamine,Gln)、甘氨酸(glycine,Gly)、脯氨酸(proline,Pro)、苯丙氨酸(phenylalanine,Phe)、异亮氨酸(isoleucine,Ile)、亮氨酸(leucine,Leu)含量有显著变化(P<0.05);作用48 h,细胞中有12种氨基酸:谷氨酸、谷氨酰胺、甘氨酸、脯氨酸、苯丙氨酸、异亮氨酸、亮氨酸、丝氨酸(serine,Ser)、组氨酸(histidine,His)、赖氨酸(lysine,Lys)、丙氨酸(alanine,Ala)、色氨酸(tryptophan,Trp)含量有显著变化(P<0.01)。 结论:细胞活力无显著差异时,细胞氨基酸代谢已有明显变化,氨基酸代谢异常可能为细胞损伤早期的一种特征。

Abstract: Objective: The main objective of this article is to estimating the effect of dimethyl sulfoxide (DMSO) on livability and amino acid metabolism of SH-SY5Y cells. Methods: SH-SY5Y cells were divided into normal group and DMSO group. The DMSO group was exposed to 0.0125% DMSO in different time (24, 48 h) to investigate amino acid transform which was associated with cell damage.SH-SY5Y cell livability was determined by MTT as well as amino acids content was determined by high performance liquid chromatography. Results: Compared with the normal group, DMSO group`s 7 amino acids (glutamic acid, glutamine, glycine, proline, phenylalanine, isoleucine, leucine) decreased significantly after SH-SY5Y cells were treated with DMSO for 24 h (P<0.05),and 12 amino acids (glutamic acid, glutamine, glycine, proline, phenylalanine, isoleucine, leucine, serine, histidine, lysine, alanine, tryptophan) decreased significantly after treated with DMSO for 48 h (P<0.01). But there was no significant transform in cell vitality and morphological specificity. Conclusions: Although there was no obvious change in cell vitality and morphological specificity, but amino acid concentrations had significant transform.  This finding suggests that abnormal amino acid metabolism may be a potential early and causal feature of cell damage.
